An excellent question, and one that comes up a lot for designers working with spot color. There are several ways to make sure your spot color jobs preview properly in Adobe Acrobat.

Sometimes the box appears slightly transparent (not 100% white.) To remedy both instances I make a white box (on a new layer if you want) and set the blending option to multiply (in the Effects palette.) This forces InDesign to render the whole page and not just the drop shadow part. Presto, no more white/translucent box in the resulting pdf! Prints good too.
